Niacinamide, the broad spectrum anti-ageing miracle

Niacinamide is a water-soluble vitamin and part of the vitamin B group. It is known as vitamin B3 which is essential in human diet. Niacinamide was first recognised as vitamin PP in the early 20th century due to its capability in preventing pellegra, an epidemic disease with severe cutaneous lesions.

The development of Niacinamide as a topical anti-ageing miracle, was pioneered by two scientists, Feldmann, RJ and Maibach, HI (Journal of Investigative Dermatology) which highlighted the good skin penetration of Niacinamide. Unlike some other anti-ageing ingredients, Niacinamide is a broad-spectrum topical anti-ageing:

1. Niacinamide improves skin moisturisation and soothes sensitive skin

A study performed by Tanno et al (British Journal of Dermatology) showed that Niacinamide reduced transepidermal water loss in stratum corneum* by 24% in 4 weeks. At the same time, Niacinamide boosted free fatty acids and ceramides in stratum corneum* by 67% and 34%.

Extended exposure to sun light is a main reason for hyperpigmentation. Melanocytes in deeper skin layers produce melanosomes that contain the pigment melanin.

A study performed by Hakozaki, T et al (British Journal of Dermatology) showed that Niacinamide inhibits the transfer of the melanosomes to the surrounding keratinocytes up to 68%. Vitamin C is an essential nutrient in our daily diet. As a potent antioxidant, vitamin C protects our body from oxidative stress. Vitamin C is also a key factor in collagen synthesis and a healthy level of collagen is the key to youthfulness. It is no surprise that vitamin C is often added into anti-ageing skincare products. However, due to the volatility of vitamin C, many skincare products with vitamin C do not deliver their promises.

The lack of stability of vitamin C in skincare products has been an issue from the very beginning. Contrary to its role as a potent antioxidant, pure vitamin C is easily oxidised in water. The oxidised vitamin C simply doesn’t provide any benefit. Luckily, science has come up with the next generation of vitamin C which is stable, known as vitamin C derivative or vitamin C ester.

The question is how to know whether a product is formulated using a stable vitamin C or not? Below are commonly used vitamin C and vitamin C derivatives:

1. L-Ascorbic acid (synonym Ascorbic acid):
L-ascorbic acid is a pure, water-soluble vitamin C. To be effective, L-ascorbic acid has to be present at a high concentration and at low ph (acidic formula). Skincare product(s) with a low ph can be too irritating for sensitive skin, in addition to that, L-ascorbic acid is very volatile, easily oxidised in water. Therefore L-Ascorbic acid is least favourable vitamin C in skincare product. To prevent a reaction with water, some companies make vitamin C skincare products using L-Ascorbic acid in a silicone-based formula (cyclomethicone, dimethicone, dimethicone crosspolymer, polysilicone-11 to name a few) instead of a water-based formula, however the efficacy of L-Ascorbic acid in a silicone-based formula is questionable. Silicone creates a ‘plastic film’ on the skin which gives an artificial, non-greasy smooth-feeling, however, it also clogs the pores so L-Ascorbic acid can’t penetrate the skin.

Nowadays, L-Ascorbic acid is widely used in food and cosmetic industry as a ph adjuster/modifier, however, the use of L-Ascorbic acid as a source of vitamin C in skincare products is no longer favourable.

2. Sodium Ascorbyl Phosphate (SAP)
Sodium Ascorbyl Phosphate (SAP) is a stable, water-soluble vitamin C derivative. Laboratory tests and studies in America, Europe and Asia supported the efficacy of Sodium Ascorbyl Phosphate:
2.1. SAP provides the amazing properties of vitamin C in a gentle form (skin-friendly), with a similar ph to pure water, it is suitable for all skin types including sensitive, fragile skin around the eyes.
2.2. SAP promotes the synthesis of collagen I and III in human fibroblasts. As we all know that healthy level of collagen is the key to youthfulness (firm and lineless skin), SAP is an excellent topical anti-ageing ingredient.
2.3. SAP evens out skin-tone and complexion. SAP reduces melanogenesis (the formation of melanin) by up to 57%. In Japan, a country with very strict regulation, SAP has been approved as a quasi-drug for skin brightening/lightening (treatment for hyperpigmentation).
2.4. SAP in combination with Tocopheryl Acetate (Vitamin E) boost the protection level of a sunblock/sunscreen. Photo-damage of skin including premature skin ageing is to a large extent caused by reactive oxygen species (ROS). A dramatic increase of ROS often result in significant damage to cell structures (oxidative stress). A combination of Sodium Ascorbyl Phosphate (SAP) and Tocopheryl Acetate (Vitamin E) provides an additional quenching effect of ROS (by up to 47%) to a standard sunscreen.
2.5. A study performed at a university in Europe, proved the efficacy of SAP in the prevention and treatment of acne vulgaris. Another independent study performed by a team from various universities and a hospital in Japan, showed that SAP improves the appearance of skin conditions associated with pimples and inflammation. During a 12 weeks study, 75.7% of 37 patients with 10 to 50 inflammatory lesions and 10 to 100 non-inflammatory lesions, experienced either good or excellent improvement with twice-daily application of SAP lotion.

Sodium Ascorbyl Phosphate (SAP) is my favourite source of vitamin C for Henry Tianus Advanced Natural Cosmeceuticals as its efficacy and safety are very well established. 3. Magnesium Ascorbyl Phosphate (MAP)
Magnesium Ascorbyl Phosphate (MAP) is a stable, water-soluble vitamin C derivative. MAP does provide similar benefits to Sodium Ascorbyl Phosphate (SAP). However, due to its molecule structure, which is easily recrytallised (at a high concentration), Magnesium Ascorbyl Phosphate (MAP) is often used at a lower concentration.

4. Ascorbyl Palmitate
Ascorbyl Palmitate is a stable, oil-soluble vitamin C derivative. Some studies showed that Ascorbyl Palmitate is less effective than Sodium Ascorbyl Phosphate and Magnesium Ascorbyl Phosphate. This may be due to its oil-solubility. By nature, vitamin C is water-soluble, forcing it to become oil-soluble is like forcing a human being to live under the water. This may also be the reason why a silicon-based vitamin C product is not as effective as a water-based one.

5. Other vitamin C derivatives
There are other vitamin C derivatives such as Ascorbyl Glucoside, Tetrahexyldecyl Ascorbate, to name a few. However with limited researches/studies to back up their efficacy, it is hard to know how effective these relatively new vitamin C derivatives are.

Worth knowing:

TRUE or FALSE
I. The easiest way to spot the oxidised vitamin C skincare product is by looking at the colour, either yellow or brown?

FALSE
Many products are formulated with other ingredients that have strong colours, for example: Retinol Palmitate (Vitamin A), Rosehip oil or botanical extracts have strong colours that influence the colour of the skincare product.

When buying a vitamin C skincare product, it is best to read the label and ingredient list thoroughly to:

1. Look for a stabilised vitamin C derivative, either Sodium Ascorbyl Phosphate or Magnesium Ascorbyl Phosphate.
As a general rule, ingredients are listed from the highest to the lowest concentration of the finish product, therefore if this active ingredient is listed close to the bottom of the ingredient list, the product may not work (due to a low concentration of active ingredient).

2. Check the expiry date. Most of active ingredients have a limited effective-life including stable, vitamin C derivatives. A skincare product can be strongly preserved, either naturally or chemically to have a long shelf-life (well past 2.5 years), therefore under current regulation, the manufacturer doesn’t have to print the expiry date. However, if the active ingredient is already passed its effective-life, this skincare product will not work effectively. Choose a product with an expiry date printed on the bottle/box!

II. A product formulated with few types of vitamin C is more effective than a product formulated with one type of vitamin C

FALSE
A product formulated with 10% Sodium Ascorbyl Phosphate is more effective than a product formulated with 3.5% Magnesium Ascorbyl Phosphate, 3.5% L-Ascorbic acid, 3% Ascorbyl Palmitate.
A product formulated with a stable, effective source of vitamin C (either Sodium Ascorbyl Phosphate or Magnesium Ascorbyl Phosphate) is more effective than a product with many but less stable/less effective sources of vitamin C.

Anti-ageing skincare: what really works? (Part 2 - Vitamin A and its derivatives)

For decades, vitamin A and its derivatives have been used extensively to reverse the signs of premature ageing skin. In term of their potency as “anti-ageing ingredients“, there are basically 3 forms of vitamin A and its derivatives:

1. Tretinoin (synonym: Retinoic acid)
The first and by far is the most active form of vitamin A, Tretinoin is an acid form of vitamin A. Topical use of Tretinoin has been clinically tested and proven to help:
- reducing the appearance of wrinkles and fine lines
- stimulating the collagen production
- improving skin texture (smoothness, age spots/pigmentation, acne)

Background:
For decades, Tretinoin (synonym: Retinoic acid) has been used by doctors/dermatologists to treat acne skin conditions. However some patients reported that the treated skin appears to be smoother and wrinkles, fine lines and age spots appear to be less noticeable. Therefore for many years, it has also been prescribed to reduce the signs of premature ageing skin.

Efficacy:
- Analysis of pre and post treated skin in a double blind study at the University of Pennsylvania's School of Medicine, Philadelphia confirmed the efficacy of Tretinoin in reversing the signs of premature ageing skin.
- Some other universities have conducted some sort of studies and also confirmed the efficacy of Tretinoin in reversing the signs of premature ageing skin due to sun exposure.

Pros:
- Has been tested clinically and used extensively to reverse the signs of premature ageing skin due to sun exposure
- Has been used for decades in treating acne and proven to be effective
- Only one application (at night time) is needed within 24 hours

Cons:
- Can be very irritating. Many people experience redness, scaling, itching and burning sensation when using a product containing Tretinoin, therefore Tretinoin is not suitable for sensitive and dry skin.
- A product containing Tretinoin increases the risk of extreme sunburn and general skin sensitivity, therefore post-treated skin should not be exposed to ultraviolet light (sun, sun lamps, tanning beds), cold winds and hair-removal products (wax, cream, laser etc.) .
- Due to associated side effects, in many countries, a product containing Tretinoin is only available through prescription.

2. Retinol
Retinol is a potent form of fat-soluble vitamin A. Although it is less “active” compared to Tretinoin, a study at the University of Michigan in Ann Arbor confirmed that Retinol can reduce the appearance of wrinkles and fine lines. Retinol possesses less side-effects than Tretinoin, however, those with sensitive and dry skin should be careful when using a product containing Retinol.

Products containing Retinol are available without prescription, but due to instability of Retinol in skincare formulas and possible side effects, many skincare manufacturers prefer to use Retinol Palmitate, a vitamin A ester, instead of Retinol.

3. Retinol Palmitate (synonym Retinyl Palmitate)
Retinol Palmitate is a vitamin A ester, arguably the most stable form of vitamin A used in skincare formulas. It is no surprise that Retinol Palmitate (synonym Retinyl Palmitate) is listed as one of the ingredients in many anti-ageing creams, lotions and potions including those from premium, organic-certified brands. Unlike Tretinoin and Retinol, Retinol Palmitate is suitable for all skin types (including those with sensitive and dry skin). In foods industry, it is often added to milk and other dairy products. Retinol Palmitate (synonym Retinyl Palmitate) is also commonly used as a vitamin supplement for treatment of vitamin A deficiency.

Worth knowing:
It is important to know that vitamin A plays important role in:
- regulating our body immune system which helps prevent or fight off infections
- maintaining healthy vision, bone growth, reproduction, cell division, and cell differentiation

Foods that containing vitamin A include liver (chicken or beef), milk, cheese, egg, carrot, kale, cantaloupe, spinach, mango, etc. The recommended daily intake for adults (RDA) is between 2,310 IU for women (slightly higher for pregnant women) and 3,000 IU for men. However extreme dose of vitamin A on daily basis is not recommended, in fact The California Environmental Protection Agency (CAL/EPA) has identified vitamin A and its derivatives as developmental toxins when administered at doses greater than 10,000 International Units (IU)*.

Skincare products containing vitamin A and its derivatives have been subjected to negative publicity associated with a relation between excessive daily intake of vitamin A (greater than 10,000 IU) during pregnancy and the increasing risk of birth defects. Although I have not known any case to prove that daily use of skincare products containing vitamin A and its derivatives contributed to any birth defects, as a precautious action and for peace in mind, any pregnant or maybe pregnant women or who are seeking to be pregnant, should inform their GP (doctor) if they use any skincare product containing vitamin A and its derivative, so their GP (doctor) can take this into consideration when recommending prenatal supplements that often contain higher doses of vitamin A than regular daily supplements.

I have been formulating skincare products (initially for my own use, family and friends) for more than two decades. Since at the very beginning, it has been my top priority to choose only safe, effective, high quality ingredients for my product range. For this reason, I decided to use Retinol Palmitate (synonym Retinyl Palmitate), the most stable, gentle and safe form of vitamin A, as one of ingredients.

Anti-ageing skincare: what really works? (Part 1 - Hexapeptide 11)

I get a lot of enquiries whether natural, organic creams, lotions and potions are better than high-tech, doctor’ brands or the opposite?

Personally I believe that “ unless the product is made using very best ingredients, it will not deliver very best result “. Natural, organic ingredients are my favourites, however my top priority is to find anti-ageing ingredients that work. Without these active ingredients, any anti-ageing skincare product is not going to work, as simple as that.

Hexapeptide 11

Background:
Hexapeptide 11 is enzymatically made using fermentation biotechnology from Saccharomyces cerevisiae, a species of yeast with well-documented wound healing properties. It increases the cellular oxygen consumption, improves the collagen and increases blood vessel development.

Research and Development of Hexapeptide 11 involved:
- Identification of the peptide fragment at the University of California, Davis Molecular Structure Facility
- Employing the powerful sequencing tool BLAST2® (Washington University)

Efficacy:
In vitro data – Gene Microarray Data:
In vitro Gene (DNA) Microarray Analysis done on human dermal fibroblasts indicates that Hexapeptide 11 influences a number of human skin genes related to stress and extracellular matrix function. In addition, Hexapeptide 11 has a powerful influence on a number of skin growth factors (EGF - Epidermal Growth Factor and FGF - Fibroblast Growth Factor).

In vivo data – Skin Firmness:
In vivo half-face study on 25 volunteers with topical twice daily application of toner containing Hexapeptide 11 around the eyes and cheeks within 4 weeks (28 days) showed that there is an improvement in the elastic response of the skin. Hexapeptide 11 helps to firm the skin compared to placebos. No allergic reaction is found.

Pros:
1. Unlike some other peptides, Hexapeptide 11 is from natural origin (Saccharomyces cerevisiae, a species of yeast).
2. Tested using biotechnology (Gene -DNA- Microarray Analysis) and direct skin application (Half-face study). Both showed positive results.
3. Firms up the skin without paralysing or “ relaxing ” the muscles.
4. Hypoallergenic (Do not normally cause allergic reaction)

Cons:
None.
